Thin-layer chromatography of amino acids and their derivatives
PTH-amino acids
Thin-Layer Chromatography on silica gel has proved highly useful for the identification of phenylthiohydantoin derivatives of Amino Acids (PTH-amino acids). For the tentative identification of these compounds, Brenner et al. [4] recommended two-dimensional Separation, first in a chloroform–methanol (9:1) mixture and then in a chloroform–formic acid (100:5) mixture. To identify the PTH-amino acids, standard solutions of known Amino acids are fractionated on the same chromatographic plate.
Chromatography of PTH-Amino acids can also be carried out in other Solvents. The spots formed by the amino acids are visualized with chlorotolidine reagent.
