Slide 104. Fundus of the Stomach.
Staining: hematoxylin and Congo red.
Magnification: x80.
Examine the slide with the naked eye, noting its distinctive dark-yellow color. Under low magnification, identify depressions on the mucosal surface known as gastric pits (1), which extend deeper into the gastric glands (2). Beneath the Base of the glands, bundles of smooth myocytes (muscular lamina of the mucosa) are located (3). Deeper still lies the submucosa (4), containing nerve and vascular submucosal plexuses.
The Superficial layer of the mucosa consists of a simple columnar secretory epithelium (5). The epithelial Cells feature a transparent, pale pink Cytoplasm. The Glands of the gastric fundus appear as simple, straight tubules. Under high magnification, the glandular wall is composed of chief exocrine cells (cells with basophilic cytoplasm) (6), parietal (oxyphilic/acidophilic) exocrine cells (large cells with oxyphilic cytoplasm) (7), neck and mucous neck cells (8), and endocrine cells. Loose Connective Tissue, representing the lamina propria of the mucosa, is found between the glands (9).
The muscularis externa consists of three layers of smooth Muscle. The serosa (Peritoneum) is a thin connective tissue layer covered by mesothelium.
