Protein Chromatography on a Cation-Exchange Cellulose Column
Fractionation of Blood Serum Proteins on a CM-Cellulose Column

A. Column preparation. The CM-Cellulose column is prepared in the same manner as the P-cellulose column, but washing of the ion exchanger differs slightly; it is performed as follows: 0.5 M NaOH, distilled Water, 0.1 M acetic acid, distilled water, and starting buffer solution.

B. Chromatography. Fractionation is carried out in the same manner as on the P-cellulose column. The following Buffer solutions are recommended for successive elution steps: 0.02 M acetate buffer, pH 4.6 (starting), 0.05 M acetate buffer, pH 5.2, 0.08 M acetate buffer, pH 6.0, 0.1 M phosphate buffer, pH 7.0, and 0.1 M phosphate buffer + 0.5 M NaCl, pH 9.2.

NOTES

1. The optimal column size and The amount of protein to be loaded for a given cation exchanger preparation must be determined in preliminary experiments in each case.

2. During regeneration, cation-exchange celluloses are treated in the same manner as during column preparation for chromatography.
