Introduction

Microbiological examinations occupy a pivotal place in the comprehensive suite of clinical and Laboratory tests used for the Prevention, Diagnosis, and Treatment of infectious diseases. Modern clinical medicine places high demands on microscopic examinations, requiring both an increased volume of tests and enhanced diagnostic quality.

Due to insufficient data on many microorganisms, contemporary classifications largely fail to reflect their phylogenetic relationships, serving instead as a practical guide to navigate the vast number and Morphological diversity of forms. The morphological characteristics and tinctorial properties of microorganisms are considered core Classification criteria. This tutorial is dedicated to the Methods used for studying them.

The Morphology and cellular Structure of microorganisms—whose dimensions are typically measured in microns (1 μ = 0.001 mm = 10-3 mm)—are studied using specialized instruments called microscopes, which provide magnifications ranging from hundreds (light microscopes) to tens of thousands (electron microscopes) of times. To reveal specific morphological features (such as cell shape, arrangement, size, motility, spore- and capsule-forming capacity, and intracellular inclusions), quantify microbial populations, check culture purity, and fulfill various other objectives, researchers prepare fixed, stained specimens that can be stored for extended periods. Chapter 5 outlines simple and differential staining methods.
