Phosphonates
Fosfomycin is a broad-spectrum antibiotic active against strains of Staphylococcus (including methicillin-resistant strains), Streptococcus, N. gonorrhoeae, N. meningitidis, E. coli, S. marcescens, Salmonella, Shigella, P. aeruginosa, H. influenzae, and V. cholerae. It is less active against indole-producing strains of Proteus and Klebsiella.
Fosfomycin readily crosses the Blood-Brain barrier and penetrates Bone tissue and Bile, with two-thirds of the dose excreted by the Kidneys. The daily dose of 3 g is divided into 3–4 administrations, with a Treatment duration of 5–7 days.
It is classified as a reserve antibiotic and is used to treat meningitis and Sepsis caused by Escherichia coli, staphylococci, and Serratia (in combination with Other Antibiotics); Osteomyelitis; cholecystitis, salmonellosis, and Helicobacter-induced enteritis; and Urinary Tract infections. When combined with beta-lactam antibiotics and Aminoglycosides, synergistic antibacterial effects are observed.
Side effects: nausea and diarrhea, which resolve upon discontinuation of the drug.
