Cleavage of Disulfide Bonds

For the accurate Identification of disulfide-linked Peptides, it is necessary to cleave the Disulfide Bonds and isolate the resulting fragments.

Methods for cleaving disulfide bonds are discussed in Sections 1.5.1 and 2.2. The standard approaches are performic acid oxidation [41] and reduction followed by alkylation [48]. The resulting half-cystine peptides are isolated using conventional Protein Chemistry techniques, including Gel filtration, Chromatography, paper Electrophoresis, High-Performance Liquid Chromatography, and others.
