Micro-scale amino acid sequence analysis using a gas-phase peptide-protein sequencer
Sequencer
The design of the gas-phase Sequencer is described in detail in [4]. Below, only certain Design Features of the instrument and its operation are noted (Fig. 17.1). A commercial version of the sequencer exists (see the ed. note at the end of the chapter), which differs from the prototype in the following ways:
1) electrical (instead of pneumatic) actuation of the Valves supplying Reagents and Solvents;
2) microprocessor control of the instrument (instead of a hard-wired cycle programmer) with the capability of modifying reaction parameters in individual cycles (for example, Cleavage time can be increased in cycles where Proline residues are expected to be present).
