Analysis programs vary depending on the instrument, Solvents, and buffers used during the FITC coupling stage. Before operation, the packed reaction Column and reagent supply tubing are washed with all solvents to be employed (for example, methanol and 1,2-dichloroethane). In a dual-column Sequencer setup, analysis on the first column begins by washing it with methanol, followed by the Condensation reaction (delivery of buffer and FITC solution). At the same time, the second column is washed with 1,2-dichloroethane, followed by a TFA wash. If necessary, the second column can be used as an automatic converter. The thiazolinone transferred to the second column after the first cycle is converted to PTH while simultaneously carrying out the carbamoylation stage of the second cycle on the first column (see below). The microanalysis program is shown in Fig. 12.5, and its adaptation for the LKB sequencer is described in Table 12.2.
