TFA and HFBA are used in Edman Amino Acid Sequence analysis and are available in a highly purified state. Buffers based on these acids are volatile and transparent at 215 nm. The Use of TFA and HFBA for peptide Separation has been described [1, 30]. These acids are ideal as Solvents for peptide isolation; a typical example is the fractionation of a tryptic digest of cytochrome c on a Vydac C18 Column using an acetonitrile concentration gradient (Fig. 6.10).
