Isolation of peptides using phosphoric acid
HPLC was used to monitor the Enzymatic Cleavage of the Calcitonin-binding protein [15]. The hydrolyzate was applied to the Column without prior Treatment. Peptide fractionation was performed in 0.1% phosphoric acid (pH 2.2) using an acetonitrile concentration gradient. During the preparative Isolation of the Peptides, the optical absorbance of the eluate was monitored at 210, 254, and 280 nm. The phosphoric acid remaining after fraction lyophilization did not interfere with Amino acid analysis, the determination of N-terminal Amino Acids using dansyl chloride, or automated amino acid sequencing. The enzymatic hydrolyzate was separated without preliminary treatment.
