The ratio of different monomers (or polypeptide chains) in an oligomer can be calculated from amino acid analysis data. This method is particularly effective if the oligomer and monomers differ significantly in their content of specific Amino Acids [29, 185].

For an oligomer consisting of two subunits, α and β, the content of any amino acid is calculated using the formula:

where X, Xa and Xβ are The amino acid content (in mole percent) in the oligomer and subunits, respectively, and a and b are coefficients reflecting the mass ratio of the a and ß subunits. Regression analysis is used to determine the coefficients a and b.
