Chapter 16. DNA VACCINES

16.2.GENE SELECTION

The Selection of genes for DNA vaccination can be performed by constructing a Gene library (for example, using the Polymerase Chain Reaction with random oligonucleotide primers), including cloning of restriction fragments of the pathogen's genome in various reading frames and sequential screening of the resulting recombinant Plasmids that provide protective Immunity. This makes it possible to identify a specific gene whose Proteins protect the Organism against infection. In cases where the protective protein is already known, its gene is obtained using the polymerase chain reaction with specific oligonucleotide primers.

Finally, to broaden the antigenic spectrum of DNA Vaccines, vectors have been developed that incorporate cDNA libraries of specific pathogens. Inoculation with a pool of such vectors expressing the library after immunization successfully protected mice against pathogen challenge, even when the specific protective Antigens were unknown.

Genes used for the construction of DNA vaccines must be conserved.
