4. METHOD FOR DETERMINING HEMOGLOBIN USING THE SAHLI HEMOMETER
The method is based on comparing the color of the test Blood solution with a standard. The Sahli hemometer (Fig. 4.3) consists of a stand with three test tubes: the two outer, sealed tubes contain a standard solution of acid hematin (16.67%).

Using a capillary, add 0.1 N HCl to the middle graduated test tube up to the lower mark. Then add 0.02 ml of blood using the capillary. Mix the contents with a Glass rod and leave for 4–5 minutes until Hemoglobin is converted into acid hematin (the solution color will change from red to brown).
Add distilled Water drop by drop using a pipette until the color of the solution matches that of the standard tubes.
Record the solution level in the middle graduated tube.
This value represents the hemoglobin concentration in the blood.
