Among the formed elements, erythrocytes (1) constitute the vast majority. They are shaped like biconcave discs; their central region is thinner and thus stains more lightly, whereas the peripheral zone is stained red or deep pink by eosin.
Among granulocytes, neutrophilic granulocytes (2) are the most frequently observed. These are Cells with a transparent, pale pink Cytoplasm and fine granules. The latter can only be discerned in well-stained preparations using an oil immersion objective. Segmented neutrophils feature a dark purple, segmented Nucleus consisting of 2 to 5 lobes. Occasionally, band neutrophils with an S-shaped or horseshoe-shaped nucleus can be identified in the specimen. Immature forms with a Kidney-shaped (bean-shaped) nucleus are very rarely encountered. Eosinophilic granulocytes (3) are less common; their nucleus typically possesses two segments, and the cytoplasm is packed with large red granules. Basophilic granulocytes (4) contain a pale, poorly segmented nucleus composed of two lobes, along with dark purple granules of varying sizes.
Lymphocytes (5) are agranulocytes characterized by a large, round, dark purple nucleus and a narrow rim of basophilic cytoplasm. Monocytes (6) are the largest leukocytes in size, surrounded by the nuclei of fibrocytes (2), featuring a kidney-shaped nucleus and a light blue cytoplasm.
Platelets are approximately three times smaller than erythrocytes, appear in small clusters, and exhibit a pale purple stain.
