Slide 26. Mesothelium (Simple Squamous Epithelium).

Staining: silver nitrate impregnation and hematoxylin. Magnification: x400.

The slide is a whole-mount preparation of the greater omentum treated with silver nitrate to delineate the borders of the epithelial Cells. The color of the preparation ranges from yellow to dark brown. The peritoneal mesothelial cells

(1) - flat epithelial cells, or mesotheliocytes - are viewed from the surface rather than in cross-section, revealing their polygonal shape. Their borders

(2) appear as jagged or wavy black lines. The Cytoplasm of the mesotheliocytes sometimes contains small silver granules. One or two nuclei (3) are stained blue or purple with hematoxylin. Blood Vessels (4) surrounded by fat cells can also be observed in the preparation.
