Mixed Anhydride Method - Mixed Anhydrides with Carbonic Acid Monoesters

Anhydrides of carbonic acid monoesters play an exceptional role in Peptide Synthesis compared to mixed anhydrides of other types. This method was simultaneously proposed by Wieland, Bernhard, and Boissonnas. The main advantage and merit of this method is the absence of Side Reactions during the peptide bond formation stage. Upon aminolysis (reaction with the amino component) of the mixed anhydride, apart from the corresponding peptide, only carbon dioxide and alcohol are formed:

Mixed anhydrides with carbonic acid monoesters are usually prepared at low temperatures (0°C and below). The peptide bond formation reaction proceeds rapidly even at room Temperature. In some cases, heating to a boil is required to complete the reaction. The process proceeds satisfactorily with the majority of Amino Acids.
