Most LDL appears to be formed from VLDL, as described above, although there is evidence that a certain fraction of LDL is synthesized directly in the Liver. The half-life of the LDL-associated apolipoprotein B in the bloodstream is approximately 2.5 days.
Studies conducted on cultures of human fibroblasts, lymphocytes, and arterial smooth Muscle Cells have demonstrated that these cells possess specific binding sites, or receptors, for LDL (B-100 receptors), which are dysfunctional in familial hypercholesterolemia. Approximately 50% of LDL degradation occurs in extrahepatic Tissues, while the remaining 50% takes place in the liver. There is a direct correlation between the likelihood of developing coronary atherosclerosis and the concentration of serum LDL.
