The amount of peptide required to determine its Structure by mass spectrometry is determined mainly by the size and polarity of the compound and its fragmentation behavior. When using instruments whose sensitivity is comparable to that of the KRATOS MS-50 or VG Analytical ZAB mass spectrometers, 30—50 nmol of a decapeptide is generally sufficient for a single analysis. However, for reliable Interpretation of mass spectra of complex mixtures, it is almost always necessary to carry out parallel experiments with deuterated Peptides (Section 19.6.3), which doubles the amount of substance required for the analysis.
