Application of electron impact mass spectrometry for determining the amino acid sequence of peptides and proteins
Requirements for the analyzed peptides
Molecular dimensions
The length of Peptides that can be analyzed by Electron Impact Mass Spectrometry is determined primarily by their volatility, and typically the number of amino acid residues in such compounds does not exceed ten. In the case of peptides built predominantly from residues of small hydrophobic Amino Acids (Glycine, valine, Alanine), twenty-residue compounds may also prove sufficiently volatile. Larger peptides can also be analyzed mass-spectrometrically, but only if they can thermally dissociate in the ion source into smaller, highly volatile fragments whose Amino Acid Sequence is determined from the mass spectrum.
