Since the mass spectrometric sequencing strategy described here is designed for peptide mixtures, the tolerance for peptide impurities in the sample is no stricter than in the dansyl-Edman method for Primary Structure determination. The advantage of the mass spectrometric method, however, is that along with determining the Amino Acid Sequence of the target peptide(s), it is often possible to establish the Introduction/19.html">Primary structure of minor components as well. It should be borne in mind that the presence of non-peptide-like impurities—such as Parafilm components, salts, grease residues, and plasticizers—is strictly unacceptable. For example, trace amounts of organosilicon compounds from silicone grease almost completely suppress the electron-impact ionization of Peptides.
