Concentration of Protein Solutions Using Sephadex
Principle of the method. Due to its high hydrophilicity, dry Sephadex added to the solution to be concentrated rapidly absorbs Water, thereby concentrating the high-molecular-weight compounds remaining in the solution.
Applications. Concentration of chromatographic fractions, Proteins isolated by other Fractionation Methods, biological fluids, etc.
PROCEDURE
Dry Sephadex G-25 (coarse) is added to the solution to be concentrated, the suspension is thoroughly mixed and, after 10 min, centrifuged or filtered. Each gram of dry Sephadex G-25 absorbs approximately 2.5 g of water. Following Sephadex Treatment, the resulting filtrate or supernatant becomes more concentrated than the initial solution, while its pH and Ionic strength remain virtually unchanged.
NOTES
1. Performing the described procedure once allows the solution to be concentrated approximately 3-fold. If further concentration is required, the process is repeated with a fresh portion of dry Sephadex.
2. Washed and dried Sephadex can be reused for concentrating solutions.
