Staining: osmic acid.

Osmic acid stains the myelin sheath (1) of isolated nerve fibers (2) black. Inside the nerve fiber, a pale-staining strip can be distinguished – the axon (3). At regular intervals along the length of the nerve fiber, narrow oblique clefts – myelin clefts (Schmidt-Lantermann clefts) (4) – and constrictions – nodes of Ranvier (nodes of myelin sheath disruption) (5) – can be observed. Surrounding the myelin sheath is the neurolemma (6), formed by the Cytoplasm and nuclei of Schwann Cells.
