Slide 30. Non-keratinized stratified squamous epithelium of the cornea.
Staining: hematoxylin and eosin.
Magnification: x400.
The section appears as a narrow pink strip. Under low magnification, orient the slide so that the wide band of the anterior epithelium is at the top, and the narrow band (posterior epithelium) is at the bottom.
Examine the wide band under high magnification. This is a non-keratinized stratified squamous epithelium. It is separated from the underlying Connective Tissue (1) by a basement membrane (2). Resting on the basement membrane is the basal layer of prismatic Cells with oval nuclei (3). Above this layer lie the parabasal and intermediate layers (4), containing several rows of polygonal cells with large, rounded nuclei. The most superficial zone shows several rows of elongated nuclei oriented parallel to the surface, which belong to flat cells (5). This is the superficial layer.
