Staining: iron hematoxylin.

Magnification: x400.

Spermatozoa are examined using a stained smear. Under low magnification, select an area with a sparse distribution of spermatozoa and switch the Microscope to high magnification. In a spermatozoon, the HEAD (1) and tail are clearly outlined. The head contains The Nucleus (2) and a narrow rim of Cytoplasm with a cap (3) at the anterior end. In the tail, we can distinguish the connecting piece or neck (4), the middle piece (5), as well as the principal (6) and end (7) pieces.
