Materials
Coupling Buffers
Buffer No. 1. N-Methylmorpholine – TFA, pH 9.5. Mix 5 mL of N-methylmorpholine and 5 mL of Water; the solution pH is 11.4. Add 4 drops of TFA to bring the pH to 9.5. Use freshly prepared buffer and store under nitrogen.
Buffer No. 2. Pyridine – Hydrochloric acid, pH 5.0. To 80 mL of water, add 8 mL of pyridine, followed by 5.3 mL of 32% hydrochloric acid, and bring the final volume to 100 mL with water (pH 5.0). Store under nitrogen.
Buffer No. 3 (anhydrous). DMF – triethylamine. To 200 mL of DMF, add 2 mL of triethylamine. Coupling is also performed in anhydrous DMF, but without The addition of triethylamine (see carboxyl coupling).
Buffer No. 4. Bicarbonate, pH 9.0. 0.1 M sodium bicarbonate is adjusted to pH 9.0 with 1 M sodium hydroxide; the buffer contains 10% (v/v) 2-propanolamine.
