Traditional strategy for protein structure determination
Fractionation of insoluble peptides
Insoluble (core) Peptides can be solubilized and then separated under denaturing conditions, but it is generally more advantageous to carry out their preliminary fragmentation. Elastase or Thermolysin is used for the Hydrolysis of poorly soluble aliphatic peptide mixtures, while Chymotrypsin or Pepsin at acidic pH is used for aromatic peptides. The resulting fragments are separated by paper Chromatography or, if the core constitutes a significant portion of the protein molecule, on a Dowex-50 Column with final purification of the obtained fractions on paper.
