Internal Standards

An accurately known amount of an amino acid—an internal standard, the recovery of which serves as the basis for calculating the yields of protein Amino Acids—is added to the protein solution prior to Hydrolysis. When using a two-Column method, different standards are required. The choice of a standard compound is determined by the individual characteristics of each column and, to some extent, by the preferences and habits of the researcher. In any case, one should strive to find the optimal option. For the long column, norleucine, norvaline, or nitrotyrosine can be tried, while for the short column (used for basic amino acids), diaminopimelic acid or homoarginine can be used [314].
