Columns packed with sorbent having a particle diameter of 5–10 µm and 10 nm pores are suitable for the Separation of A wide variety of Peptides, but for the isolation of high molecular weight Polypeptides (≥20,000), it is more advisable to use sorbents with pore diameters of ≥50 nm. The Use of acidic buffers with high salt concentrations prevents the interaction of polypeptides with unmodified silanol groups. Some columns (Ultrapore RPSC by Altex, Vydac, Zorbax C8, Bioseries by Dupont) feature a high degree of additional modification performed to eliminate residual silanol groups, making these columns preferable for peptide isolation. Currently, sorbents coated with C3, C4, and C8 Hydrocarbons are the most effective. Particularly good separation is achieved using the Ultrapore RPSC (C3) Column (Fig. 6.9).
