2. METHODOLOGY FOR LEUKOCYTE COUNTING
Dilute the Blood 20-fold with a 3% acetic acid solution containing gentian violet. This can be done in two ways: first, draw blood into a leukocyte pipette (Fig. 4.5) up to the 0.5 mark, and then draw in the acetic acid solution up to the 10.1 mark; second, add 0.4 ml of the gentian violet-stained 3% acetic acid solution into a clean, dry test tube, and then add 0.02 ml of blood using a micropipette.

Place a drop of the diluted blood onto a Goryaev chamber prepared in the same manner as for erythrocyte counting. Count the leukocytes under low magnification in 100 large squares grouped in sets of 4, using a darkened field of view.
