If ∆H0 can be assumed constant over the temperature range of the experiment, a plot of lnC versus 1/T makes it possible to determine ∆H0 (or ∆H' if the plot is constructed for lnK'): the slope of the line is equal to —∆H0/R. Since knowing K allows one to calculate ∆G0, equation (3-14) can then be used to determine ∆S0. However, this method yields low accuracy, and therefore it is preferable to measure ∆H directly by calorimetry. Moreover, the assumption that ∆H0 remains constant over a wide temperature range, particularly for Proteins, may prove to be incorrect.
