Опыты на пептидных гомополимерах позволили установить, что некоторые аминокистотные остатки обнаруживают свойство встраиваться в а-Спираль. Эти данные были затем применены к глобулярным белкам с известным пространственным строением. Оказалось, что остатки, которые образуют спирали в гомополимерах, стремятся встраиваться в спирали также и в глобулярных белках. Такое соответствие послужило основой для многочисленных попыток установить корреляции между аминокислотной последовательностью белка и наличием спиралей в этом белке. Позднее такие корреляции были распространены и на другие вторичные структуры. Эти попытки интересны с той точки зрения, что они являются некой основой пока еще не известного будущего метода, с помощью которого можно будет устанавливать трехмерную структуру белка только по его аминокислотной последовательности. Это и послужило причиной подробного описания большинства из существующих методов «предсказания» вторичной структуры по аминокислотной последовательности. Current Methods make it possible to predict the correct Secondary Structure for approximately two thirds of all protein residues. There are no major differences in prediction quality between Probabilistic methods based solely on the mathematical analysis of structural data and Physicochemical methods that also incorporate additional stereochemical information. This indicates that our understanding of the physical basis of these correlations is still insufficient. Until recently, comparing different prediction methods was rather difficult because authors used different performance metrics. The relationship between these metrics and their relative merits are discussed in this book. Such a comparative evaluation of methods can contribute to their further development. A reliable prediction method can prove useful not only in determining three-dimensional structures, but may also provide valuable insight into the folding process by identifying the chain segments that fold first.
