The degradation cycle According to the DABITC–PITC method is shown schematically in Fig. 14.1. It should be noted that

1) DABITC is not very stable in pyridine, and its solution is best prepared daily or prior to each degradation cycle;

2) some hydrophobic Peptides tend to precipitate as fine particles or a viscous film during extraction. The precipitate typically concentrates at the interface between the aqueous and organic phases after centrifugation and may be inadvertently removed with a pipette.
