Methods of solid-phase amino acid sequence analysis

Reagents
Methyl isothiocyanate (MITC) (Fluka, C.P. grade) and phenyl isothiocyanate (PITC) (Fluka, C.P. grade) were vacuum-distilled (oil pump) under nitrogen, and the fraction boiling at 4 °C was collected. They were stored in small portions in ampoules at —20 °C.
p-Phenylene diisothiocyanate (DITC) (Eastman). If the reagent is yellowish, it is recrystallized from boiling acetone: 2 g of DITC is dissolved in 10 mL of acetone and cooled slowly. Yield: 1.55 g (needle-like crystals yielding a clear, colorless solution in DMF). tпл 129—131 °С.
4-N,N-Dimethylaminoazobenzene-4'-isothiocyanate (DABITC) (Fluka or Pierce). Recrystallized from acetone: 1 g of DABITC is dissolved in 70 mL of boiling acetone, filtered through filter paper, and cooled slowly. Yield: ~0.7 g of orange-red or brown needle-like crystals. tnл 169—170 °С.
1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC) (Serva). tпл 112—115 °С.
3-Aminopropyltriethoxysilane (Pierce)
N-β-Aminoethyl-(3-aminopropyl)trimethoxysilane (Pierce).
